ChrisRay over the the Nvidia forums has put together a great and very in-depth Nvidia multi GPU SLI/PhysX setup guide. Starting with the typical routine driver installation required on any new setup ChrisRay then goes in-depth on the different SLI/PhysX configurations you can run and how to setup them up. He then shows several performance examples using Batman Arkham Asylum to show how the different setups will benefit from having a dedicated PhysX card. If you have a multi-gpu/SLI or just two Nvidia video cards and need to figure out how to setup dedicated PhysX then you will definitely want to check out this guide.

Welcome: With the vast amount of SLI configurations and PhysX Setups available. It can be a little confusing on how to setup your system and peak it for optimal performance. This is a general guide to understanding how Nvidia Multi GPU and PhysX processing works. This guide will be divided into 3 Parts.
1) SLI Setup And Installation for 2way/3way/Quad SLI
2) SLI Profiling System And AFR explanations
3) PhysX Processing in Multi GPU Enviroment And Setup
Introduction: With the introduction of PhysX. A whole new way to use your GPUS beyond traditional SLI rendering has come about. Here we will go over turning PhysX on, Setting PhysX devices, and how individual SLI and multi GPU configurations will work as a PhysX rendering setup.
Enable GPU PhysX: In order to utilize GPU PhysX. The driver must be turned on to have it enabled. To do this simply enter the SLI and PhysX tab and select enable PhysX. This will turn your GPU into a PhysX device. If you do not have this enabled. All GPU PhysX games will fall back to CPU PhysX.

Now this is exciting news, Epic Games has announced in their GDC 2012 press release that they will be showcasing Unreal Engine 4 behind closed doors at GDC 2012.
